I have access to a Lux amp 15114454 out of a denali....

Would I have to do any progamming changes to it for a Silverodo 2500 Crew cab to sound perfect?

IE set it up as a Escalade EXT?

I do have access to a GM TechII and Shop.

you don't have to change anything - should sound fine as is (at least mine did with the same amp). You can reprogram for the EXT and it will improve some.

At least I would know right away if I got things wired and working right away then..... That is good at least.

Is there a guide or step by step on reprogramming the LUX Amps with a Tech II?

Does the EXT have separate tweets in the rear or is it exactly the same lay out?

There's a guide in the DIY post on the topic, it's a link that shows the instructions as I remember. I don't know if the EXT has the 2nd tweeters but it won't hurt your sound either way. Pretty straight forward doing it with the tech 2, just drill down through the Body controls to the AMP and it's a special funtion as I remember.

I have an 08 LTZ crew Cab with the Bose 6disc, would the Lux be a plug and play??

The Lux amp used in the Classic body style won't work in yours - don't know about lux amps for the NBS, if there are lux amps for them, or if they need to be rewired to work.
